BCIP and NBT

Reagents for the detection of alkaline phosphatase in biochemical assays.

BCIP (5-Bromo-4-chloro-3-indolyl-phosphate p-toluidin salt)
A substrate of the alkaline phosphatase, which releases the blue stain 5-Bromo-4-chloro-3-indoxyl by cleaving the substrate. The enzymatically released indoxyl will be oxidized by NBT to reveal the insoluble indigo.

NBT (Nitro blue tetrazolium chloride)
Determination of the activity of alkaline phosphatase in combination with BCIP. This reaction is based on the oxidation of indoxyl to the insoluble, blue indigo by NBT.
